PSA algorithm for digital readers

The Panel Score Algorithm (PSA) calculates a single numerical value from signals of individual analytes (biomarkers) measured by a digital reader. The numerical PSA value is compared to a Cutoff value for the Panel Score used to interpret the test results. Positive test has PSA>Cutoff and Negative test has PSA≤Cutoff. This technology is patent pending: U.S. Continuation-In-Part Application entitled "Systems and Methods for Analyzing Persistent Homeostatic Perturbations”. US PTO No: 15/148,968. TCA algorithm for Saliva Image Analysis

The Tri Partite Classification (TCA) algorithm is the first available tool for quantitative analysis of biomarkers in whole saliva. The algorithm is applied to a database of digitized saliva images. The images are produced by a laboratory instrument for analyzing biological samples, or a digital reader. The algorithm is implemented using automated high troughput procedure. Results provide total signal volume (TSV) for biomarker expression level stratified for epithelial cells, leukocytes and fluid saliva. TSV is used to calculate biomarker data, and determine the biomarker distribution between saliva cells and fluid.

  1. Standard fields of whole saliva on a microscopy slide were stained using sandwich immunoassay with red label. The slide was digitized using a scanning microscopy. Field #4 (green frame) was selected for image analysis.
  2. Red label bound to Mucin 1 protein.
  3. Image analysis using TCA algorithm. The digital image was segmented: red mask-epithelial cells, blue mask-leukocytes, cyan mask-saliva fluid.
  4. Staining intensity - white mask.

SRP algorithm for Pathway Analysis

The Stress Response Profile (SRP) algorithm calculates Pathway signature from biomarker data. The algorithm is applied to a database of biomarkers data produced by a digital reader or a laboratory instrument for analyzing biological samples. The SRP algorithm calculates Pathway Activation Index for each pathway. The numerical value of the Index indicates the pathway activation level. Preferentially activated pathways have the greatest Z value. Pathway analysis provides new insights into the molecular mechanism of disease that can be translated into innovative diagnostic tests or treatments. This technology is protected by U.S. Patent No. 8,518,649 “Systems and Methods for Analyzing Persistent Homeostatic Perturbations”.

Pathway signatures of disease: comparison of acute and chronic phase. TBI, Traumatic brain injury. NCD, neuroicognitive disorder. HF-pEF and HF-rEF, acute heart failure with preserved or restricted ejection function. Acute HIV, seroconversion. HIV/AIDS, AIDS patients not responsive to antiretroviral therapy. The Pathway activation index was calculated from normalized biomarker data using a patented algorithm, and normalized. Arrow indicates the preferentially activated pathway. SRP pathways: 1-Redox, 2-Detox, 3-Chaperoning, 4-DNA, 5-Adhesion/Cytoskeleton, 6-Cell cycle, 7-Apoptosis, 8-Signaling, 9-Immunity, 10-Microbiome.
